What is ACE-031?

ACE-031 (ramatercept) is a soluble fusion protein constructed from the extracellular domain of the activin type IIB receptor joined to an immunoglobulin Fc fragment. Rather than blocking a receptor on the cell surface, it functions as a circulating decoy: it binds ligands of the TGF-beta superfamily — myostatin (GDF-8) and related activins among them — before they can engage their native receptors. Because myostatin is the principal negative regulator of skeletal muscle mass, sequestering it is a clean way to interrogate that braking mechanism. The Fc domain provides the extended stability characteristic of fusion-protein constructs.
